CRISPR/CAS9 BASED DNA-COMBING ASSAY FOR DETECTING ANTIMICROBIAL RESISTANCE GENES ON PLASMIDS

We present a method based on CRISPR/Cas9 excision and DNA combing to detect anti-microbial resistance (AMR) genes on bacterial plasmids. The assay is inexpensive, simple, fast, and also provides information on the number and size of plasmids in a sample. We demonstrate detection of the gene encoding for the New Delhi metallobeta-lactamase 1 (blaNDM-1) enzyme, known to make bacteria resistant to a

Inertia induced breakdown of acoustic sorting efficiency at high flow rates

We investigated the impact of inertial forces on acoustic sorting at high sample throughputs. Above a critical flow rate, we observed an unexpected inertia induced breakdown of the sorting efficiency that is not due to the wall induced lift force and was not previously described in the literature. We analyzed the effect experimentally and theoretically, providing an explanation to the phenomena, a

Entomological Scheimpflug lidar for estimating unique insect classes in-situ field test from Ivory Coast

Acquisition of entomological data with high-frequency lidar is an emerging research field in rapid development. The technique offers very high numbers of observations per time unit, suitable for statistical models. In this work, we use a near-infrared Scheimpflug lidar with a sampling frequency of 3.5 kHz to assess the activity of free flying organisms. In-situ measurements were done during the ra

Acoustically Excited Channel Walls for Microbiological Applications

This work presents a novel concept for acoustofluidic cell manipulation: thin vibrating channel walls. Improved etch recipes enable us to produce thin walls between two microfluidic channels, fabricated in silicon or via PDMS casting, that can be used for challenging microbiological applications. We demonstrate the medium exchange for bacteria utilised for transformation, single-cell rotation for

Glances and hands on public art : A kaleidoscopic view of monuments in public space in the Öresund region and beyond

How do we relate to monuments and statues in public spaces? And how do our perceptions shift when history confronts us with new insights? In the wake of the Black Lives Matter movement in 2020, many monuments and statues across the world became the focal point of intense debate. Some were torn down, others covered up, and others were re-examined in light of not only colonial and racial oppression,

Schartau och herrnhutismen: Erfarenheten, själavårdsbreven, efterverkningarna

Tunable separation and DNA manipulation in metal coated pillar arrays

Implementing electrically connected metal-coated posts in a Deterministic Lateral Displacement (DLD) device and applying electric fields, we use electrokinetics to achieve tunable particle separations and to trap and manipulate DNA. The strength of DLD stems from its typically binary behavior. Particles move in one out of two trajectories based on their effective size. For particles that are close

ACouWash : A standalone instrument for the washing, separation and enrichment of cells.

We developed the AcouWash (Figures 1 and 2), a benchtop instrument for flow-through cell and particle separation by acoustophoresis. Building on previous research [1-4] AcouWash can re-suspend cells in new medium, separate cells based on size or acoustic properties, or enrich cells, Figure 3. We present our initial evaluation of AcouWash regarding size-based separation of particles, cell washing,

Cerebral extracellular calcium activity in severe hypoglycemia : Relation to extracellular potassium and energy state

The changes in extracellular Ca2+ (Cae) and K+ (Ke) activities were studied in the rat brain during insulin-induced hypoglycemia. At about the time of onset of isoelectric EEG in severe insulin-induced hypoglycemia (300-g male Wistar rats under 70% N2O anaesthesia), there was an increase in Ke which, at ∼13 mM, was associated with a fall in Cae. Ke peaked at 48 ± 12 mM, and Cae at 0.18 ± 0.28 mM.

Subjectivism and Relational Good

In this paper, a distinctly subjectivist analysis of the nature of relational goodness or goodness for is proposed. Like the generic subjectivist analysis of value, the proposal is to analyse value in terms of attitudes. Specifically, the proposed analysis of goodness for appeals to a special kind of attitude: namely, so-called for-someone’s-sake attitudes. Unlike other analyses in the literature

Value of Information analysis accounting for data quality

Structural Health Monitoring (SHM) can provide valuable information for maintenance-related activities and post-disaster emergency management. However, as with any technological system, SHM systems can be susceptible to errors due to malfunctioning. Therefore, it is essential to assess the potential for malfunctions and the associated costs of maintenance and repair when evaluating the long-term b

SEPARATION OF SINGLETS AND CLUSTERS OF GROUP A STREPTOCOCCI USING DETERMINISTIC LATERAL DISPLACEMENT AND FILTER SONICATION

Differences in morphologies of bacteria and bacteria clusters are thought to contribute to their virulence and colonization. However, the conventional standard cell biological methods cannot separate bacteria and bacteria clusters based on their morphologies and sizes, making studies of the underlying mechanisms difficult. Here we report a simple label-free method for the continuous separation of

Artificial life goes 'in silico'

The paper reviews a number of cellular neural network implementations on a field-programmable gate-array. It illustrates experimentally, how these implementations can be used to measure from images or to create dynamic, autonomous processes that facilitate measurements within topographic maps. Applications range from image understanding to robot navigation

Toward low-carbon ways of life : The cultural politics of contesting aeromobility

Min aThe need for demand-side climate solutions is increasingly recognized in science and society. Such solutions imply that demand for goods and services is targeted either via changes in the use of infrastructure and technology or via cultural changes in consumption and lifestyle among the global affluent. In this thesis, I explore how to enable cultural change around affluent overconsumption through

Helium identification with LHCb

The identification of helium nuclei at LHCb is achieved using a method based on measurements of ionisation losses in the silicon sensors and timing measurements in the Outer Tracker drift tubes. The background from photon conversions is reduced using the RICH detectors and an isolation requirement. The method is developed using pp collision data at √s = 13 TeV recorded by the LHCb experiment in th
