Levodopa treatment provides marked symptomatic relief. However, within 5-10 years after the start of Levodopa treatment, most PD patients display a gradual loss of drug efficacy. Importantly, the neuropathology of PD is dominated by a marked loss of dopaminergic neurons located in the substantia nigra.
